Cloning of genomic DNA encoding murine poly(A)-binding protein Ⅱ생쥐 Poly(A) 결합 단백질 Ⅱ 유전자의 클로닝

Formation of the 3``-end is an essential step in almost all eukaryotic messenger RNA syntheses. The genomic DNA clone for poly(A)-binding protein II, which plays a crucial role in poly(A) addition and its length control, was isolated from mouse genomic library using cDNA probe. In order to elucidate the genomic structure of this clone, named mPAB II (murine poly(A)-binding protein II), the total sequence of 8.2 kb was determined. Its sequence shows that the genome of mPAB II consists of 7 exons and 6 introns, and indicates strong homology with that of the bovine PAB II. The predicted amino acid sequence contains one putative RNA-binding domain of the RNP type. Based on the examination of sequence elements at promoter region and CpG islands profile, it is estimated that this housekeeping gene might have a TATA$^{-}$Inr$^{+}$ promoter.
