dc.description.abstractFat tissues mainly regulate metabolism and reproduction. Adipocytes generate throughout life, with the massive expansion occurring during the postnatal period. Recently, adipose stromal vascular fraction (SVF)-derived white adipocyte progenitor cells are indentified, but the identity of bone marrow-derived white and brown adipocyte progenitor cells in vivo are unknown. Moreover, adipose SVF is not only containing adipocyte progenitors, but also a mixture of blood endothelial cells, immune cells, fibroblasts, pericytes, stromal cells and yet undefined stem/progenitor cells. Previous studies have shown that mesenchymal stem/progenitor cells in SVF can be cultured to successfully differentiate into adipocytes, pericytes, osteoblasts, chondrocytes and myocytes under optimized condi-tions. However, little is known about freshly isolated donor adipose SVF can readily provide blood endothelial cells to create a vascular network through a ‘disassembly and reassembly’ mechanism. Hence, we attempt to describe the existence of bone marrow-derived circulating adipose progenitor cells, and the cellular events occurring in the process of vascular network formation from freshly isolated adipose SVF as a source candidate for cell therapy and suggest their potential therapeutic applications. Here, I divided this thesis into two chapters as follow: bone marrow-derived circulating progenitor cells fail to transdifferntiate into adipocytes in adult adipose tissues in mice (part I), and freshly isolated stromal vascular fraction from adipose tissue forms profound vascular network through the disassembly and reassembly of blood endothelial cells in mice (part II).
