We herein developed a novel isothermal method to detect platelet-derived growth factor (PDGF) utilizing target-induced nicking and extension chain reaction system-based amplification (NESBA). In principle, the binding of PDGF to the aptamer strand (AS) that was priorly hybridized with template strand (TS) liberates TS which then initiates the following NESBA system, leading to the exponential amplification of RNA amplicons that are monitored by molecular beacon (MB). With this method, we sensitively determined a model target, PDGF-BB, down to 6.48 nM with high selectivity against non-specific analytes including different types of PDGFs. More importantly, we also demonstrated the capability of this sensor to reliably analyze PDGF-BB in the human serum sample, verifying its practical applicability to the development of point-of-care (POC) device for cancer diagnosis.
