Many of preprotein containing signal sequence are known to be translocated posttanslationally across membrane. Precursor ribose binding protein from E. coli SP114 was purified and posttranslational translocation of pRBP into inverted E. coli plasma membrane vesicles was studied. Intact pRBP was not translocated across INV possibly because pRBP was not in atranslocationcompetent conformation. When pRBP dissolved in 8 M-urea solution was diluted into translocation mixture containing INV plus either ATP or NADH, posttranslational translocation across inner membrane vesicle was observed.
