Recently, chemical industry have looked for alternative energy source as depletion of fossil fuel. Shale gas is rising alternative energy source that has over 70 percentage of methane gas. Methane gas could convert to methanol by treating simple process under normal condition and methanol is attractive substrate for biotechnology application as C1 carbon source renewable feedstock. Traditionally, some bacteria have utilized methanol as main carbon source called methylotroph. However, these methylotrophs have some drawback that are lack of genetic tool and platform to synthesis of valuable product. For overcoming these disadvantages, well-known industrial bacteria Corynebacterium glutamicum is selected as construction de novo pathway. Through metabolic engineering and enzyme engineering, C. glutamicum could consume methanol as a carbon source. The impressive difference between previous research is the pathway is constructed as linear pathway instead of cycle pathway so that the methanol consumption could be performed simply and more fast. The result showed some methanol was converted as a energy source in C. glutamicum. For further optimal goal of this research is possible to utilize methanol as only carbon source in C. glutamicum, improving those induced enzymes and synthesis of high value added product.
