dc.description.abstract | Most of nanoparticles for cancer therapy is cleared in liver without reaching target cells because of phagocytosis of macrophages. Attention to nanoparticles which are not uptaken by macrophages has been uprising to overcome this limitation. However, still large proportion of injected nanoparticles is cleared by macrophages, and complete avoidance of macrophage is very challenging. In this study, we researched the possibility to compensate the efficiency of delivery inducing macrophages to secrete uptaken cargo of nanoparticle, packaged in exosomes. We tried to increase the amount of re-excreted cargo by adding ceramide family lipid, which is known to induce cells to secrete exosomes, to nanoparticle liposome composition. We showed that ceramide family lipid contained in liposome enhanced the exosome secretion of macrophage and increased the amount of liposome cargo in exosome. Furthermore, we also showed that the increase of secreted material via exosomes can result in the enhancement of delivery to tumor cells. | - |
