Quantitative and label-free investigation of microorganisms using optical diffraction tomography광 회절 단층촬영 기술을 활용한 미생물의 정량적 및 비침습적 분석

Optical diffraction tomography (ODT) is a technique for quantitatively measuring the three-dimensional (3D) refractive index distribution of a sample. Based on a quantitative phase imaging (QPI) technique, the two-dimensional (2D) optical fields are measured at various angles to reconstruct the three-dimensional refractive index distribution. Becasue the QPI technique quantitatively measurement of the optical field through an interferometer, the high contrast quantitative images of the transparaent samples can be obtained non-invasively through the phase information. In particular, the ODT have the potential to be utilized in various biological studies as the technique provide qauntitative information about the physical and biochemical characteristics as well as the morphological information of live biological samples. In this thesis, we introduce non-invasive quantitative measurement of 3D images of living biological samples using ODT, and quantitative investigation of the shape and contents of internal cellular materials. We focus on the optical measurement and quantitative analysis of internal substances of microorganisms that accumulate the biosynthesized products in the body. Although existing other imaging techniques have difficulty in quantitative live cell imaging of the biosynthesizing microorganisms, ODT enables to nondestructively reconstruct the 3D RI distribution of individual living cells. In particular, we investigate microalgae that produce lipids and bacteria that produce biodegradable plastics. We characterize the size, cell growth and accumulation rate from the reconstructed 3D refractive index distribution, and also examine the correlation between various characteristics. Real-time 3D imaging and qauntitatvie invesitagtion of living single cells is available with unique capabilities of ODT, which is label-free and quantitative 3D imaging. The capability allows to study the mechanisms underlying accumulation process of biomaterials or alteration of the cells due to the external environment. We expect that the present approach provides unprecedented oppertuninty to study biophysical processes happened in various microorganisms via non-invasive quantitative 3D imaging.
