The present invention relates to a kind of novel customizations of gene expression in reduction prokaryotes to synthesize sRNA, preparation method and its application, more particularly to a kind of synthesis sRNA, it includes the Hfq binding site from sRNA any in MicC, SgrS and MicF and the region with target gene mRNA at base-pair, further relates to the preparation method and applications of synthesis sRNA.The advantages of synthesis sRNA of the invention, is, the inhibition level of target gene can be controlled by the binding ability of regulation synthesis sRNA and target gene mRNA.Using the synthesis sRNA of regulation expression of target gene, it can effectively construct recombinant microorganism without traditional gene delection method and can reduce the expression of target gene, thus it is very useful to the production of recombinant microorganism to synthesize sRNA.In addition, synthesis sRNA can quickly be applied to various bacterial strains, thus it is very suitable to the metabolic capability and the most suitable bacterial strain of selection of measurement bacterial strain.In addition, by manipulating that metabolic flux obtains using synthesis sRNA and efficiently producing the recombinant microorganism of tyrosine or cadaverine medical highly useful with industrial circle.In other words, selection expression can be made to become easy being suppressed with the target gene for being used to efficiently produce metabolite using sRNA of the invention.Therefore, synthesis sRNA can be used for constructing the recombinant bacterial strain for effectively producing various metabolites and for establishing the effective ways for producing various metabolites, thus highly useful.
