Ｒａｂ５를 이용한 세포내 단백질 상호 작용 시각화 방법A visualizing method for monitoring intracellular protein-protein interaction using Rab５
본 발명은 엔도좀에 결합하는 Rab5 단백질을 이용하여, 살아있는 세포 내 엔도좀에서 단백질 간 상호 작용을 관찰할 수 있는 단백질 상호 결합 모니터링 방법에 관한 것으로서, 보다 구체적으로 칼슘 신호 의존적인 칼모듈린과 칼모듈린 키나아제인 CaMKIIα(calcium/calmodulin-dependent protein kinase IIα)와 CaMKK1(calcium/calmodulin-dependent protein kinase kinase 1)의 상호 작용을 본 발명의 ECLIPSE(Emerging CircLe of Interactive Proteins at Specific Endosomes) 방법으로 관찰하여, 칼모듈린과 CaMKIIα의 가역적인 상호 작용을 밝혀냈으며, 칼모듈린 키나아제인 CaMKK1과 CaMKIIα의 칼슘 신호에 대한 민감도 차이로 인하여 칼모듈린과 결합하는데 차이를 갖는 것을 확인함으로써, 본 발명의 ECLIPSE 방법이 단백질 간 상호 작용을 모니터링하는데 유용한 방법임을 확인하였다.
