심장-얼굴-피부 증후군의 유도-만능 줄기세포 모델 및 이의 용도cardiofaciocutaneous syndrome induced pluripotent stem cell model and use thereof
본 발명은 심장-얼굴-피부 증후군(cardiofaciocutaneous syndrome, CFC 신드롬)의 유도-만능 줄기세포(induced pluripotent stem cells; iPS) 모델, 이의 제조 방법, 및 상기 iPS 모델을 이용하는 CFC 신드롬의 신경학적 발달 분석에 이용하는 용도에 관한 것으로, 구체적으로 CFC 신드롬 환자의 섬유아세포로부터 CFC 신드롬 유래의 iPS 및, 배상체의 발생 및 분화를 유도하였으며, 상기 CFC 신드롬 유래의 iPS 및 배상체는 깨어진 배상체 형태를 나타내고, 신경세포로의 분화능을 나타내지 않는 것을 확인하였으며, p-ERK 및 p-SMAD1 억제제를 처리하여 CFC 신드롬 유래의 배상체를 유도한 결과, 상기 배상체는 정상적인 배상체의 형태를 나타내며, 신경세포로 효과적으로 분화하므로, 본 발명의 CFC 신드롬 환자 유래의 줄기 세포 모델은 심장-얼굴-피부 증후군의 신경 발달을 위한 분석 연구에 유용하게 사용될 수 있다.
