| dc.description.abstract | 본 발명은 부탄올 생성능을 가지는 재조합 미생물 및 이를 이용한 부탄올의 제조방법에 관한 것으로서, 더욱 상세하게는 n-butyrate를 기질로 하여 부탄올을 제조하는 재조합 미생물 및 이를 이용한 부탄올의 제조방법에 관한 것이다. 본 발명에 따른 재조합 미생물은 (a) 지방산 대사경로를 가지는 미생물에서, 부티릴-CoA를 아세틸-CoA로 전환시키는 효소를 코딩하는 유전자 및 지방산 대사에 관여하는 유전자의 발현을 조절하는 효소를 코딩하는 유전자가 결실; (b) 지방산을 아실-CoA로 전환시키는 효소를 코딩하는 유전자의 attenuator를 포함하는 native promoter가 strong promoter로 치환; 및 (c) 부티레이트로부터 부탄올을 생합성하는데 관여하는 효소를 코딩하는 유전자가 도입되어 있는 것을 특징으로 한다. 본 발명에 따른 재조합 미생물은 기존의 Clostridium acetobutylicum과 달리 성장이 용이하고, 추가 대사흐름 조작으로 인한 균주개량의 가능성이 크므로 부탄올의 산업적 생성 미생물로 유용하다. | - |
