In this thesis, general-purpose readout platforms are studied in detection of various biomolecules with dielectric modulated field effect transistor (DMFET) array for point-of-care testing (POCT) applications. For preliminary study on field effect transistor (FET)-based electrical biosensors, a DNA sensor array is fabricated and characterized with an integrated current readout circuitry. The operational principle of the fabricated DNA sensor is based on the ion sensitive field effect transistor (ISFET) and the intrinsic charge of DNA affects the surface potential of the sensing region in the sensors. Various concentrations of polymerase chain reaction (PCR) amplicon are successfully quantified with the implemented readout system. As a first version of DMFET array in this research, a single-gate dielectric modulated field effect transistor (SGDMFET) array is fabricated and characterized with a custom designed readout platform. A prostate cancer marker, prostate specific antigen (PSA), and its antibody are immobilized in the vertical nanogap of SGDMFET. With the implemented readout platform, drain currents of SGDMFET array are monitored and successfully detect PSA and antibody by current increments caused by dielectric constant changes. Finally, a double-gate dielectric modulated field effect transistor (DGDMFET) array is fabricated and characterized with a revised version of readout platform. In order to verify nanogap filling with sample solution and evaporation of the sample solution from the nanogap, calculations and simulations are performed. Antigen and antibody of avian influenza are successfully detected by threshold voltage shifts caused by dielectric constant changes in the nanogap.
