핵자기공명분광법을 이용한 Telomere DNA 구조 연구Study on the telomeric DNA structure by NMR spectroscopy
telomere DNA의 말단에 해당하는 oligonucleotide들의 구조를 핵자기공명분광법을 이용하여 결정하였다. Oxitricha telomere의 외가닥 부위에 해당하는 $d(G_4T_4G_4)$ monomer의 immino peak이 12~14ppm에서 관찰되었으며, immino proton과 non-labile aromatic proton 간의 NOE 현상도 관찰되었다. 이러한 결과들로부터 monomer의 구조는 GG reverse-Hoogsteen base-pairing에 의한 hairpin 구조임을 알 수 있었다. 연결고리의 길이에 따라 안정성의 차이는 있었으나, Tetrahymena, 사람의 telomere 염기배열을 가지는 oligonucleotide들에서도 hairpin구조의 immino peak들이 관찰되었다. GG hairpin은 생체내 telomere의 기능에 잘 부합되는 구조이다.
