Study on transcription regulation by chromatin remodeler Ino80크로마틴 리모델러 Ino80의 전사 조절에 관한 연구

Chromatin remodelers are known as organizers of genome structure via nucleosome assembly, eviction, and exchange. Recent study has revealed those chromatin remodelers are also engaged in early transcription, elongation, and termination processes. I performed PRO-seq to investigate transcriptional roles of Ino80, one of the chromatin remodeler family. As a known fact, several mutants which has defects in transcription elongation and termination tends to show cold temperature sensitivity. By screening various chromatin remodeler subunit deleted strains, Ino80 complex deleted strains showed significant cold sensitive phenotype. Among Ino80 complex, subunit ies6 and ino80 were chosen and by PRO-seq analysis, deletion of Ino80 complex caused defect of RNA pol II pausing in promoter proximal region and reduced of RNA pol II signal in ORF. By this investigation, I found Ino80 complex associated with RNA pol II stability in most protein coding genes, and this could be understood by chromatin remodeling activity.
