Monoclonal Antibody specific for Cl. botulinum type F toxin was prepared using the so-called hybridoma technique. Toxin was purified through two ion-exchange chromatographic steps. It was made toxoid and then injected into BALB/C mice according to immunization schedule. After boosting injection their splenocytes were isolated and fused with NS1, one of myeloma cell lines. Hybrids were selected by HAT medium, screened by ELISA technique and cloned by limiting dilution method. Again, clones were screened by ELISA one clone was chequed as powerfully positive. We named it JY101 and cultured in 50ml T-flask. Its culture supernatant showed neutralizing activity against type F toxin. And it permits no crossreaction with botulinum toxins of other types tested.
