OBJECTIVE: Obesity is a concern worldwide. Therefore, for the control of obesity and prevention of abnormal metabolism, we focused on peripheral responses of endocannabinoid signaling with Celastrol. To further define the role of Celastrol, we performed experiments and proved the results with an obese mice model. METHODS AND RESULTS: We founded Celastrol through high throughput-screening as a CB1 receptor blocker and proved that Celastrol binds to CB1 by a binding assay. RT-PCR and an immunoblotting assay revealed a decreased level of pro-inflammatory cytokines as well as cell signaling factors that up-regulate pro-inflammatory genes on Celastrol-treated monocytes. Through CB1 activation to monocytes, we proved that Celastrol exerts a suppressing influence on the reactive oxygen species (ROS) detected by flow cytometry. Moreover, Celastrol-treated obese model mice suppressed the weight gain and decreased fat size. Real-time PCR expressed reduced level of pro-inflammatory cytokines as well as cell signaling factors on the mice heart and aorta tissues. CONCLUSION: Celastrol inhibits pro-inflammatory responses as well as ROS production. These effects were due to a CB1 receptor blockade. These results occur in obese mice along with suppression of body weight gain. Celastrol, as a natural compound, may be a promising anti-obesity agent to regulate body weight and metabolic heart diseases.
